Revise your letter or material, and keep trying.

What kind of book are you writing? Does the genre fit with a particular publisher? Who publishes books you like at B&N/Borders? Find all of this out and then tailor your letter to explain why YOUR BOOK fits with THAT PUBLISHER. Suggest how it will sell and how it can be marketable and appealing to the audience you want. Be specific though, don't just say "women will love it!" When in fact it may be narrowly tailored to a specific sub-set. Most important though is don't sweat the rejections. Once you get published the only one you'll rememebr is the acceptance letter. J.K. Rowling got rejected by several pubs before one picked her up.
